Tirzepatide is one peptide with activity at GIP and GLP-1 receptors. Retatrutide is another peptide that acts at those two receptor types and also at the glucagon receptor. “Dual” and “triple” describe their receptor profiles: a product containing retatrutide can therefore have one active ingredient and three receptor targets. Tirzepatide overview; retatrutide discovery study.
What the three signals contribute
GLP-1 connects food intake with glucose-dependent insulin release and appetite regulation. This is the shared pathway behind much of the discussion of fullness and eating less.
GIP is another meal-related signal involved in insulin release. Tirzepatide brings GIP and GLP-1 receptor activity together in a single molecule. Its pharmacology is described through that combination, rather than through a mixture of separate GIP and GLP-1 products. EMA explanation.
Glucagon is the extra receptor pathway in retatrutide. In the original development work, its contribution to greater energy expenditure was studied in obese mice alongside reduced food intake associated with the other pathways. This gives the third target a distinct metabolic rationale; human energy-use claims require human measurements. Original development study.
A useful comparison consequently asks two questions: which receptor signals are present, and what the complete molecule does when studied? Counting targets answers the first. The second depends on the properties of the molecule and its clinical results.
One ingredient is different from a premixed product
A combination such as CJC-1295/Ipamorelin names two separate peptide ingredients. Tirzepatide and Retatrutide each name one complete active substance, even though that substance interacts with multiple receptor types.
For example, “Retatrutide 32 mg” describes the declared total content of that one compound. A CJC-1295/Ipamorelin product instead needs a separate content statement for each component. This makes the ingredient count visible alongside the receptor profile.
Where clinical results fit
Both compounds have been investigated in large obesity trials: SURMOUNT-1 for tirzepatide and the phase 3 TRIUMPH-1 publication for retatrutide in September 2026. Those are separate trials, not a direct randomized comparison between these two compounds. SURMOUNT-1; TRIUMPH-1.
For the molecular question, the answer is specific: retatrutide adds glucagon-receptor activity to a GIP/GLP-1 profile. For a purchase, the ingredient name then needs to be connected to the actual finished product; a studied molecule and a particular catalog pen are different levels of information.
